Our study focused on evaluating the antioxidant and antibacterial effect of the aqueous extract of leaves of Pistacia lentiscus L. harvested from the Shafia region (El-Tarif) and essential oil, and preparing zinc oxide nanoparticles based on them. Several analyzes were used, including HPLC/MS, GC/MS/FID, UV-vis, XRD, SEM-EDS, FT-IR. The Chemical Screening tests revealed the leaves' richness in secondary metabolite compounds, including tannins, alkaloids, polyphenols, and sterols. The results of the quantitative estimation of phenols showed that the aqueous extract contained (61.703±10.041 μg EAG/ml), and as for tannins, it contained (118.7272±2.07 μg EC /ml), and the flavonoids was (32±3.791 μg QE/g DM). As for the antioxidant activity tests, which were conducted with three different methods (DPPH, FRAP, β-carotene), revealed an antioxidant response. The results of the antiradical activity of DPPH• showed the highest inhibition values when the aqueous extract (169.61±9.35 μg EAA/mg), while the essential oil was (358.53±15.73 μg EAA/mg) and for the values of each of the zinc oxide for the aqueous extract and the zinc oxide nanoparticles for the essential oil, their values were less than IC50, so we couldn’t calculate it. The same for the FRAP, the value of the aqueous extract was estimated to be (318.091±7.2 μg/mg) and the essential oil was (138.13±8.41 μg/mg). As for the zinc oxide extracts, there was no activity after several repetitions. As well as for the β- carotene test, where the aqueous extract was able to inhibit (83.33%), and the percentage of essential oil was close to (80.68%). The antibacterial activity was evaluated on seven bacterial strains pathogenic to humans: Pseudomonas aeruginosa, Escherichia coli, Staphylococcus aureus, Staphylococcus homonis. All extracts showed similar results, But the essential oil gave the best resultsen_US
