    Etude Phytochimique et l’activité biologique de l’extrait de fruits de quelques variétés d’Abelmoschus Esculentus L

    الملخص: تهدف هذه الدراسة إلى تثمين ثمار نبات القناوية، من خلال دراسة المحتوى الفينولي و النشاط البيولوجي للمستخلص الخام لثمار نبات القناوية Abelmoschus esculentus L. لمنطقة الوادي. استعملنا ثمار مقطوفة في ثلاثة أشهر مختلفة (أوت، سبتمبر و أكتوبر) و ثلاث طرق للاستخلاص و مذيبين. تم التقدير الكمي للمركبات الفينولية باستخدام كاشف الفولين FC، تم استخدام العديد من الاختبارات الكيميائية الحيوية لتقييم خواصها المضادة للأكسدة: DPPH˙، FRAP و TACعلاوة على ذلك، تم فحص النشاط المضاد للبكتيريا باستخدام طريقة الانتشار ضد الإشريكية القولونية، المكورات العنقودية، الزائفة الزنجارية، المكورات العنقودية الذهبية، السالمونيلا التيفية و الكلبسييلا. أظهرت النتائج أن مستخلص ثمارAbelmoschus esculentus L. يحتوي على كميات مهمة من مادة البوليفينول بما في ذلك الفلافونويد و الفلافونول، و يمكن اعتبارها مصادر جيدة لهذه المركبات للتطبيقات الطبية والغذائية.كما أوضحت النتائج أن مستخلص شهر أوت أغنى بالمكونات الفيتوكيميائية كمادة الفلافانول، الفلافونيدات و متعددات الفينول حيث وصلت قيمة كل منها على التوالي(190.115.75 ملغ/غ، 24.210.54 ملغ/غ و0.059.87 ملغ/غ)، و هذا ما أعطاه أعلى نشاط مضاد للأكسدة بتثبيط الجذر الحر DPPH˙ و TACوFRAP مقارنة بمستخلص شهر سبتمبر و أكتوبر حيث قدرت قيمة IC50 بـ 14.330.55) لمستخلص أوت>15.33±0.58 لمستخلص سبتمبر> 17.430.63 ملغ/مل لمستخلص أكتوبر بالنسبة لاختبار DPPH). أظهرت جميع السلالات البكتيرية المدروسة حساسية عالية ضد مستخلص الثمار و كان لمستخلص شهر أوت أفضل نشاط مضاد للجراثيم ضد المكورات العنقودية الذهبية .و كشفت النتائج أن مستخلص الأسيتون غني بالمكونات الفيتوكيميائية و له أعلى نشاط مضاد للأكسدة. بينت نتائج تأثير طرق الاستخلاص أن طريقة الاستخلاص بجهاز سوكسلي هي الأمثل لاستخلاص المركبات الفينولية بالتالي تبدي قدرات مضادة للأكسدة عالية مقارنة بالاستخلاص بالنقع و طريقة الأمواج الفوق الصوتية. أما عن النشاط التثبيطي للبكتيريا فوجد أن السلالات البكتيرية المدروسة كانت حساسة لمختلف المستخلصات و أن لمستخلص الأسيتون أفضل نشاط مضاد للبكتيريا ضد المكورات العنقودية الذهبية و الكلبسييلا. يمكن للمركبات الفينولية أن تلعب أدوارًا مهمة في النشاط المضاد للأكسدة لـ Abelmoschus esculentus L. استنادًا إلى العلاقة الجيدة الموجودة بين النشاط و المحتويات الفيتوكيميائية و التي تشير إلى أن التأثيرات الملحوظة يمكن أن تُعزى إلى المركبات الفينولية. و عليه يمكن استخدام مستخلص ثمار Abelmoschus esculentus L. في العديد من التطبيقات في المستقبل. Résumé : فرنسية أو انجلزية The objective of this study was to determine the phytochemical profile, antioxidant, and antibacterial activity of fruit extracts from Abelmoschus esculentus L. for El-Oued region. We used three methods of extraction, fruits collected in August, September, October and two solvents. The total phenolic content evaluated in fruit extracts using Folin-Ciocalteu reagent. Several biochemical assays were used to evaluate their antioxidant properties: (DPPH, FRAP and TAC). Further, antibacterial activity was screened using disk diffusion method against Escherichia coli, Staphylococcus, Pseudomonas aerigunosa, Staphylococcus aureus, Salmonella typhi and Klepsiella. Three fruits extracts of Abelmoschus esculentus L. was found to contain a high total phenolic content (190.11± 5.75 for AFE, 159.64± 4.28 for SFE 138.19± 4.34 mg GAE/g DW for OFE). The fruit extract exhibited potent antioxidant activity determined by DPPH˙, FRAP, and total antioxidant activity assays, the high antioxidant capacity of all extracts has been observed and related to the relative amounts of total phenolic content with good antioxidant properties (DPPH˙ assay, IC50= 14.33±0.55 mg/ml for AFE> IC50= 15.33±0.58 mg/ml for SFE> IC50= 17.43±0.63 mg/ml OFE). All bacteria showed high sensitivity against fruits extract and the AFE had the best antibacterial activity against Staphylococcus aureus. The results obtained for the photochemical analysis and the DPPH˙ test gave a good result to the Soxhlet method, but the maceration method has a high antioxidant property compared to the other methods. The results revealed that acetone extract is very rich in polyphenols, flavonoids and flavonols, the same extract had the highest antioxidant activity. As for the antibacterial inhibition, it was found that the studied bacterial strains were sensitive to different extracts and the acetone extract had the best antibacterial activity against Staphylococcus aureus and Klepsiella. Phenolic compounds may play important roles in the antioxidant activity of Abelmoschus esculentus L. based on The good correlation found between activity and phytochemical contents indicates that effects observed could be attributed to phenolic compounds. The results suggest that the fruits of Abelmoschus esculentus L. can be considered as a good source of natural antioxidant and antibacterial drugs and may be used in many applications in the future.
