مسلم, أم الهناءمأمون, لبنىسبوعي, خديجة2023-01-242023-01-242022-09-21university of el ouedhttps://dspace.univ-eloued.dz/handle/123456789/14260مدكرة ماستر كيمياء"تم في هذه الدراسة ، دراسة الخصائص البنيوية و الإلكترونية و الطاقوية للنواه الأساسية لحمض الكافئيك بواسطة عدة طرق نظرية مثل DFT , Ab-initio, PM3 ثم كخطوة ثانية تم تطوير نموذجين QSARلـــــ 16 مشتقا لحمض الكافيئيك لديهم نشاط بيولوجي ضد نوعين من الباكتيريا بإستخدام طريقة الإنحدار الخطي(MLR). تم حساب الواصفات المستخدمة بوسطة طريقة DFT/B3LYP/6- 31G* لبرنامح Gaussian09، وكذلك برنامج Hyperchem، ……swissADME وتم تقييم قدرة التنبؤ لنماذج QSARبواسطة طريقة (LOO) وقد لوحظ وجود إرتباط بين قيم النشاط التجريبي والمتوقع , مما يدل على الجودة العالية لنماذج QSARالمتحصل عليها. وأخيرا تم التنبؤ بالحركية الدوائية للمركبات أي تأثير الكائن الحي على الدواء خلال حركته داخل الجسم عبر أربع مراحل تعرف بــADME ، و تم ذلك عن طريق دراسة خاصية التشبه بالأدويةdruglikeness و كذلك التنبؤ بإحتمالية الإمتصاص المعوي HIAو النفادية عبر الحاجز الدموي الدماغي((BBB. ""In this study, the structural, electronic and energetic properties of the basic nucleus of caffeic acid were studied by several theoretical methods such as DFT , abinitio, PM3 In the next step, two QSAR models were developed using the MLR method for 16 caffiec acid with activity antibacterial. The descriptors used were calculated by DFT/ B3LYP/6-31G' method inGaussian09 program, Hyperchem, swissADME ……. The prediction ability of these QSAR models were evaluated by (LOO) method. A correlation was observed between the experimental and predicted activity values, which indicates the high quality of QSAR models obtained. Finally, the pharmacokinetics of the compounds were predicted, i.e. the effect of the organism on the drug during its movement inside the body through 4 stages known as ADME, and this was done by studying the drug-likeness property, as well as predicting the probability of intestinal absorption (HIA) and Permeability across the blood-brain barrier (BBB).
